Learning Outcomes and Competencies The aim of the practical is to learn the skills to plan and implement a CHO cell-based mAb production in fed-batch mode in benchtop bioreactors (well-mixed and agitated). Participants will harvest and purify the product (IgG), and present the experimental results per group in a protocol, compare them with data from the literature and also perform a cost analysis of the experiment. The experiment is carried out over a period of 14 days and is to be conducted as a block course of half a day each. For safety reasons, a maximum of 12 students per course can work in the laboratory.
Module Content
  • Inoculum production in shake flasks and in the CL 1000
  • Preparation of the bioreactors
  • Inoculation of the bioreactors
  • Feeding of the bioreactors
  • Harvesting of the bioreactors and product purification
  • Experimental data acquisition (growth and product formation curves, substrate and metabolite profiles) and troubleshooting
  • Literature search to evaluate the experimental results
  • Evaluation and logging of data
Follow-up Modules None
Methods of Instruction  Research, presentations, logging of data
Digital Resources Videos
Lesson Structure / Workload
 Contact Hours 52
 Guided Self-Study 6
 Independent Self-Study 32
 Total Workload 90
Classroom Attendance Yes
Assessment Coursework 100%: Each group (2, max. 3 students) must submit a report for grading. In the event of unexcused absence from the practical, a technical paper on the topic of the practical (approx. 10 pages) must be written instead and submitted for grading (grade must be ≥ 4).
Language of Instruction  German
Comments The module Practical Cell Culture Technology 2 is part of the Bioprocess Development and Bioengineering module group.
